Phenol/Chloroform RNA Isolation Method
A. Guanidine HCl Homogenization Buffer for RNA Isolation: 8.0M Guanidine HCl, 0.1M Sodium Acetate, pH 5.5, 5.0mM Dithiothreitol, 0.5% Sodium Lauryl Sarcosinate
B. Extraction with Phenol: Most extractions are performed using equal volumes of equilibrated phenol with respect to the sample volume. Normally, the extraction is performed twice with phenol and once with a 1:1 solution of phenol/chloroform. Hence, it is estimated that 25 ml of phenol is required for every 10 ml of sample.
Note: If regular phenol is purchased, it will need to be equilibrated before use. For this reason, many customers may prefer to purchase phenol from USB that is already equilibrated (Product Code 75829 or 77510).
